There have been lots of discussion on the subject and I am sure you 
would find quite a few resources online.
But in a nutshel, Maildir, the format supported and created by Dr. 
Bernstein, author of Qmail has the advantage of being less risky than a 
Mailbox since a Maildir holds a mail message per file. It (Qmail) 
handles nicely file locking over the NFS too. It's great for small to 
medium system. It is also quicker and safer for application to manage a 
Maildir than a Mailbox.
Mailbox is good for larger system and has the advantage over Qmail for 
those kind of big systems to consume much less inodes.
